The present study included a total of 200 throat swab were collected from patients suffering from pharyngitisthe isolates were identified according to cultural , biochemical tests , VITEK 2 system compact. Molecular detection of streptolysin O (SLO) gene by using specific primers was also studied and the results show that all isolates contain this gene with molecular length in (628 bp) Purification of Streptolysin O from crude was done by high performance liquid chromatography( HPLC ).The molecular weight of fractions was determined by using SDS-PAGE which indicates the molecular weight of fraction was estimated as 64 KD for Streptolysin O protein.
